An Aqueous Two Phase Partitioning System Of Peg Enzyme And Tannic Acid For Recovery And Reuse Of Cellulase In Palm Waste Saccharifleation from the Author: G. Dutta. The application of an aqueous two-phase partitioning system for the recovery and reuse of enzyme was studied. By addition of polyethylene glycol (PEG) to a complex of tannic acid (TA) and cellulase, a two-phase system was developed with liberation of the enzyme in solution for reuse in the saccharification of palm press fibres (PPF). Investigation of the phase separation and ensymes recovery indicated that the enzyme concentration required was 3% (v/v) which means that its concentration should be around 8% (w/w) during hydrolysis before dilution with TA and PEG. The conditions required for the formation of two-phase systems include a pH range of 4.0-5.5, temperature of 30-45 C and addition of NaCl (0.5M). The conditions conducive for maximum recovery were pH 5.5 and a temperature of 45 C. It was also observed from the results that TA may partially inhibit enzyme activity. [Authors' abstract].
